Die circadiane Uhr terrestrischer Organismen erlaubt die Anpassung von Physiologie und Verhalten an 24 h Rhythmen der Umwelt wie beispielsweise den täglichen Licht-Dunkel-Wechsel. Rhyparobia maderae wurde als Organismus zur Forschung an der inneren Uhr etabliert. Die circadiane Uhr der Madeira Schabe ist die akzessorische Medulla (AME) im optischen Lobus, welche von etwa 240 Neuronen innerviert wird. Diese Neuronen exprimieren und kolokalisieren auffällig viele Neuropeptide, die als Neuromodulatoren/Neurotransmitter des Uhrnetzwerks fungieren. Ich habe die Rolle der myoinhibitorischen Peptide (MIPs) und des ion transport peptide (ITP) in Schaltkreisen des circadianen Systems von R. maderae untersucht. Dabei habe ich mich auf Eingangsbahnen fokussiert, welche die Uhr an externe Rhythmen koppeln, ebenso wie auf Ausgangsnetzwerke, die circadiane Rhythmen der Physiologie und des Verhaltens steuern. Immunzytochemische Mehrfachfärbungen, histochemische Methoden und backfills ermöglichten die neuropeptiderge Charakterisierung von Eingangsbahnen, welche die innere Uhr an den Licht-Dunkel-Wechsel anpassen. Vermutlich wird die Lichtinformation vom Komplexauge direkt in der proximalen Lamina (kurze Photorezeptorzellen) und indirekt über MIP-immunreaktive (ir) mediale Neurone (MNes) mit Verzweigungen in den Medullaschichten ME2-4 von langen Photorezeptorzellen an zwei Ensembles von pigment-dispersing factor (PDF)-ir circadianen Schrittmacherneuronen vermittelt. Je ein Netzwerk scheint dabei als Morgen (M)- oder Abend (E)-Oszillatornetzwerk die circadiane Uhr an den Sonnenauf- oder -untergang anzukoppeln. Das größte PDF-ir Neuron empfängt vermutlich Eingänge aus dem extraokularen photorezeptiven Lamina- und Lobulaorgan, setzt PDF am Tag frei und stabilisiert antagonistisch die Phasenlage des M- zum E-Netzwerk. Injektionen von MIPs oder mip precursor (mip-pre) knockdown Experimente via RNA Interferenz in Kombination mit Verhaltensversuchen lassen die differentielle Beteiligung mehrerer MIPs an der Steuerung von M- und E-Oszillatoren vermuten. Die MIPs stabilisieren zudem Ruhe-Aktivitäts-Zyklen. Dabei kompensieren bestimmte Neuropeptide das Fehlen anderer, was für eine komplexe Modulierbarkeit des Systems durch die Freisetzung von Neuropeptiden abhängig von der Tageszeit und dem physiologischen Zustand des Tiers spricht. Auch ITP wurde in Neuronen des circadianen Netzwerks identifiziert. Eine Aufgabe von ITP in den Oszillatornetzwerken wurde aber noch nicht gefunden. Eine mip-pre knockdown abhängige Veränderung der Körpergröße deutet auf Funktionen von MIPs im Metabolismus der Weibchen während der Reproduktion. Die Ausgangsnetzwerke, die dafür verantwortlich sind, sowie die Verknüpfung der identifizierten ITP-ir neurosekretorischen Zellen mit dem circadianen System müssen noch in zukünftigen Arbeiten untersucht werden.; The circadian clock of terrestrial organisms allows the adaptation of physiology and behavior to 24 h rhythms of the environment, such as the daily light-dark cycle. Rhyparobia maderae is an established model organism for circadian research. The circadian clock of the Madeira cockroach is the accessory medulla (AME) located in each optic lobe. The AME is innervated by approximately 240 neurons. These neurons express and colocalize neuropeptides that function as neuromodulators/neurotransmitters of the circadian clock network. In my dissertation I investigated the role of myoinhibitory peptides (MIPs) and ion transport peptide (ITP) in circuits of the circadian system of R. maderae. I focused on input pathways that entrain the clock to external light-dark rhythms and output pathways that control circadian rhythms in physiology and behavior. Multiple-label immunocytochemistry, histochemistry, and backfills enabled the neuropeptidergic characterization of photic input pathways that entrain the endogenous clock to light-dark cycles. Apparently, light information is transmitted directly in the proximal lamina and indirectly via MIP-immunoreactive (ir) medial neurons (MNes) with branches in medulla layers ME2-4 from photoreceptor cells of the compound eye to two ensembles of pigment-dispersing factor (PDF)-ir circadian pacemaker neurons. Each network appears to couple the circadian clock to either sunrise or sunset as morning (M) or evening (E) oscillator circuits. The largest PDF-ir neuron is assumed to receive luminance input from the extraocular photoreceptive lamina and lobula organ, releases PDF during the day, and stabilizes the phase relationship between the M and E oscillator network through antagonistic PDF effects. Injections of MIPs or mip precursor (mip-pre) knockdown experiments via RNA interference in combination with behavioral experiments suggest the involvement of different MIPs in controlling M and E oscillators, also via stabilization of rest-activity cycles. Apparently, certain neuropeptides compensate for the absence of others, suggesting a complex modulation of the system controlled by the release of neuropeptides depending on daytime and physiological state of the animal. ITP was identified in neurons of the circadian network. However, knockdown experiments combined with locomotor assays did not elucidate its role in the circadian clock. Sex-specific modulation of body size via mip-pre knockdown experiments suggests a reproduction-dependent function of MIPs in female metabolism. The connections between the circadian system and potential output networks responsible for this effect, as well as roles of ITP-expressing neurosecretory cells, need to be investigated in the future.

All living things are subject to biological rhythms. Circadian rhythms are generated by internal clocks which influence many biochemical and physiological processes, as well as various behaviors of an organism. These oscillators are synchronized by external "Zeitgeber" signals such as light-dark changes, temperature changes, as well as social contacts. Different studies identified the accessory medulla (AME), small neuropil at the ventromedian edge of the medulla, as the circadian pacemaker center of the cockroach Rhyparobia maderae. The AME is associated with approximately 240 neurons that are categorized into seven soma groups, which express a strikingly high number of neuropeptides. The main aim of the present dissertation was to further identify and functionally characterize the neuronal circuitry of the AME of R. maderae, with a special focus on γ-aminobutyric acid (GABA)-expressing neurons. Previous studies showed that GABA-immunoreactive(-ir) fibers in the distal tract (DT) connect the AME to the medulla and about 29 GABA-ir neurons were localized near the accessory medulla. However, they never were assigned to the specific AME neurons and there were several open questions with regard to arborization patterns of GABA-ir neurons and their connections to the circadian network.
In the first part of this project I characterized the GABA-ir neurons in the brain. Regarding the circadian system, I showed that except for the anterior neurons, cells from all other soma groups of the AME expressed GABA. Next to the GABA-ir DT and the anterior fiber fan system, I discovered a new GABA-ir tract which I called medial layer fiber tract (MLFT). The GABA-ir MLFT is the third tract that strongly connected the AME to layer four of the medulla. GABA-ir fibers of the DT connect the AME to the medulla, only. I showed for the first time that the lamina organ, a putative extraretinal photoreceptor, is also GABA-ir. Regarding the optic lobe, neither photoreceptors nor lamina cells were GABA-ir, but GABA-ir medulla cells of different soma groups were found. GABA-ir neurons in the medulla might provide inhibitory feedback to the lamina and control information processing and adjust light sensitivity of the compound eyes. Additionally, basic distribution pattern of GABA immunoreactivity in the central brain provided important information for understanding the functional role of GABA in the different information processing circuitry in the brain of R. maderae.
In the second part of my thesis I was in search for a possible function of GABA and serotonin (5-HT) in the light entrainment pathway to the AME and performed multiple-label immunohistochemistry. GABA immunoreactivity in the lamina and the second layer of the medulla overlapped with terminals of the short- and long histaminergic photoreceptor neurons and. thus, could indicate possible synaptic connections in these regions. In the accessory laminae, which showed immunoreactivity to many neuropeptides, histamine immunoreactivity was absent. Additionally, I showed that GABA levels cycle in the optic lobes as well as in the DT day-time dependently. GABA-ir neurons associated with the AME showed colocalization with FMRFamide. FMRFamide showed colocalization with 5-HT and MIP in some lamina- and AME neurons. One large medium-sized PDF-ir cell colocalized with MIP and FMRFamide. For the first time, I showed that PDF lamina cells that coexpressed PDF and 5-HT projected via the anterior fiber fan from the lamina down to the AME, but not to the medulla. This pathway was the first ipsilateral input pathway from lamina neuropils that could be tracked to the AME. Double- and triple-labeled AME neurons found in the current project might indicate that the presence of different neurotransmitters in one neuron could have a cooperative function in the circadian clock.
In the third part, the role of GABA in the coupling of bilaterally symmetric accessory medullae was analyzed via backfills from the optic lobe in combination with immunohistochemistry. GABA was localized in contralateral projecting commissural medulla cells.
In the last part, in search for output areas of the circadian clock to target regions in the brain, backfill experiments form the thoracic ganglia, ocellus, and the neurohaemal organs were combined with immunohistochemistry using anti-PDF and anti-GABA antisera. I showed that large second-order- as well as 2 small multimodal ocellar neurons projected to the optic lobe and innervated the AME. The projections of the labeled ocellar neurons in the posterior optic tubercles, posterior optic commissure, lobula valley tract, and in the AME greatly overlapped with PDF and GABA immunoreactivity. I concluded that GABA might mediate ocellar entrainment of the AME. Interestingly, none of the neurosecretory cells in the brain were GABA-ir. However, the GABA-ir fibers were in close vicinity to the backfilled fibers of neurosecretory cells in the superior median- and superior lateral protocerebrum where PDF-ir fiber projections were present, which could indicate synaptic connections between the two systems. Finally, evidence was found that several regions in the brain, including the inferior lateral protocerebrum, the superior median protocerebrum, and the posterior optic tubercles which are GABA- and PDF-ir are potential premotor areas that relay circadian information to locomotor centers in the thoracic ganglia. In summary, I could contribute significantly to the characterization of the cockroach circadian clock network.

Organismen auf der Erde entwickelten oszillierende endogene Uhren, welche zirkadiane Rhythmen in der Physiologie und im Verhalten steuern, die für Gesundheit, Überleben und die Organisation von täglichen Aufgaben (z. B. Lokomotion, Nahrungsaufnahme) essentiell sind. Die endogene Periode der zirkadianen Uhren ist mit dem 24-Stunden-Licht-Dunkel-Zyklus der aufgehenden und untergehenden Sonne synchronisiert. Ebenso wie alle anderen Organismen, besitzt auch die Madeira-Schabe Rhyparobia (syn. Leucophaea) maderae eine innere Uhr mit einem endogenen Rhythmus von etwa („circa“) 24 Stunden (Golombek und Rosenstein, 2010). Läsions- und Transplantationsexperimente identifizierten die akzessorische Medulla (AME) mit pigment-dispersing factor (PDF) exprimierenden Neuronen am ventromedialen Rand der Medulla in den optischen Loben als zentrales Schrittmacherzentrum, welche den Aktiviätsrhythmus der Schaben steuert (Stengl und Homberg, 1994; Reischig und Stengl, 2003a). Die nachtaktive und robuste Madeira-Schabe ist ein gut geeigneter Modellorganismus für verhaltensbezogene, zelluläre und elektrophysiologische Untersuchungen der zirkadianen Forschung (Page 1982; Stengl und Homberg, 1994; Homberg et al., 2003). Bislang wurde die innere Uhr von Insekten, sowie auch der Madeira Schabe, nur wenig elektrophysiologisch charakterisiert. Auch ist unbekannt wie PDF-Neurone Aktivitätsrhythmen auf zellulärer Ebene steuern. In meiner Dissertation lag der Schwerpunkt auf der elektrophysiologischen Charakterisierung von AME Neuronen und im besonderen auf PDF Neuronen in vitro als auch in vivo. Das Neuropeptid PDF stellt den wichtigsten zirkadianen Kopplungsfaktor für Insekten dar. Dieses steuert vermutlich die Synchronisation von Schrittmacherneuronen beider bilateral symmetrischer zirkadianer Uhren, kontrolliert Ruhe- und Aktivitätsrhythmen und ist
am Lichteingang beteiligt (Helfrich-Foerster 2014; Stengl et al., 2015; Stengl und Arendt, 2016). Mittels intrazellulärer Ableitungen wurden in dieser Arbeit zunächst AME-Neurone abgeleitet, um zirkadiane Schrittmacherzellen, wie unter anderem auch die PDF-Zellen, weiter zu charakterisieren. Diesbezüglich wurde zunächst von mehreren lichtempfindlichen Neuronen in der Medulla oder nahe der AME abgeleitet. Dabei konnte das größte PDF-exprimierende Neuron identifiziert werden. Dieses generierte Oszillationen, reagierte tagsüber aber nicht auf applizierte Lichtreize. Da es sich als sehr schwierig erwies PDF-Neurone mit der intrazellulären Methodik vollständig zu färben und physiologisch zu charakterisieren, wurden weitere Techniken praktiziert, um PDF-Neurone weiter analysieren zu können. Bei der Durchführung von Backfills in R. maderae aus dem kontralateralen optischen Stiel zusammen mit ultrananokristallinen Diamant- (UNCD) Filmen, Calcium-Imaging- Experimenten und immunchemischen Analysen konnte gezeigt werden, dass alle PDF-sensitiven kontralateral projizierten AME-Neurone durch PDF-Applikation inhibiert wurden, während alle ipsilateral verbleibenden Neurone durch PDF aktiviert wurden. Unter den PDF-sensitiven Neuronen waren auch kontralateral projizierende mittelgroße PDF-immunreaktive (ir) Neurone, die durch PDF inhibiert wurden und kleine lokale ipsilateral verbleibende PDF-ir-Neurone, die durch PDF aktiviert wurden. Diese PDF-exprimierenden Neurone hatten Autorezeptoren für ihr eigenes Peptid. Interessanterweise besaß das größte PDF-ir-Neuron, das in allen Zielbereichen der PDF-Zellen verzweigt, keine Autorezeptoren für sein eigenes Peptid. Wie bereits zuvor in Ca2+ Imaging- Studien in Drosophila melanogaster (Liang et al., 2016) gezeigt werden konnte, variierte die elektrische Aktivität von AME-Neuronen im Laufe des Tages. Interessanterweise zeigten AME-Neurone Aktivitätsmaxima während der Abenddämmerung, bevor die lokomotorische Aktivität der nächtlichen Madeira- Schabe beginnt. Andere Zellen wurden im Morgengrauen und während des Tages aktiv, während die Schaben schlafen. Als nächstes untersuchte ich über loose patch- clamp-Ableitungen, ob Neurone in der AME auf weißes oder grünes Licht bei niedrigen Intensitäten reagieren und ob PDF lichtempfindliche Neurone in der zirkadianen Uhr der Madeira-Schabe beeinflussen kann. Wie in früheren Studien in Periplaneta Americana gezeigt wurde (Mote und Goldsmith, 1970; Mote und Black, 1981), reagierten sie speziell auf grünes Licht im Gegensatz zu weißem Licht. PDF- Applikationen verursachten Inhibitionen, Aktivierungen oder Ensemble-Formationen in der AME. Interessanterweise konnte PDF Lichtantworten von AME-Neuronen modulieren. In einigen Fällen konnte PDF auch Lichtreaktionen in der zirkadianen Uhr unterdrücken. Loose patch-clamp-Ableitungen beider AMEs zeigten, dass beide gekoppelt sind und auf die PDF-Applikation oft in gleicher Weise reagierten. Da angenommen wird, dass sowohl GABA als auch Serotonin an neuronalen Lichteingangswegen und Steuerungs-Mechanismen in der AME beteiligt sind, wurde in dieser Arbeit mit patch-clamp Ableitungen weiterhin GABA- sowie Serotonin- Reaktionen von AME-Neuronen charakterisiert. Letztlich konnte gezeigt werden, dass PDF-Neurone tatsächlich eine Rolle bei der Steuerung von Lichteingängen spielen, ebenso wie bei der kontralateralen Kopplung, der Synchronisation und Ensemblebildung in der AME, die für ihr eigenes Peptid, GABA und ACh empfindlich ist.

B. Lokomotion, Nahrungsaufnahme) essentiell sind. Die endogene Periode der zirkadianen Uhren ist mit dem 24-Stunden-Licht-Dunkel-Zyklus der aufgehenden und untergehenden Sonne synchronisiert. Ebenso wie alle anderen Organismen, besitzt auch die Madeira-Schabe Rhyparobia (syn. Leucophaea) maderae eine innere Uhr mit einem endogenen Rhythmus von etwa („circa“) 24 Stunden (Golombek und Rosenstein, 2010). Läsions- und Transplantationsexperimente identifizierten die akzessorische Medulla (AME) mit pigment-dispersing factor (PDF) exprimierenden Neuronen am ventromedialen Rand der Medulla in den optischen Loben als zentrales Schrittmacherzentrum, welche den Aktiviätsrhythmus der Schaben steuert (Stengl und Homberg, 1994; Reischig und Stengl, 2003a). Die nachtaktive und robuste Madeira-Schabe ist ein gut geeigneter Modellorganismus für verhaltensbezogene, zelluläre und elektrophysiologische Untersuchungen der zirkadianen Forschung (Page 1982; Stengl und Homberg, 1994; Homberg et al., 2003). Bislang wurde die innere Uhr von Insekten, sowie auch der Madeira Schabe, nur wenig elektrophysiologisch charakterisiert. Auch ist unbekannt wie PDF-Neurone Aktivitätsrhythmen auf zellulärer Ebene steuern. In meiner Dissertation lag der Schwerpunkt auf der elektrophysiologischen Charakterisierung von AME Neuronen und im besonderen auf PDF Neuronen in vitro als auch in vivo. Das Neuropeptid PDF stellt den wichtigsten zirkadianen Kopplungsfaktor für Insekten dar. Dieses steuert vermutlich die Synchronisation von Schrittmacherneuronen beider bilateral symmetrischer zirkadianer Uhren, kontrolliert Ruhe- und Aktivitätsrhythmen und ist
am Lichteingang beteiligt (Helfrich-Foerster 2014; Stengl et al., 2015; Stengl und Arendt, 2016). Mittels intrazellulärer Ableitungen wurden in dieser Arbeit zunächst AME-Neurone abgeleitet, um zirkadiane Schrittmacherzellen, wie unter anderem auch die PDF-Zellen, weiter zu charakterisieren. Diesbezüglich wurde zunächst von mehreren lichtempfindlichen Neuronen in der Medulla oder nahe der AME abgeleitet. Dabei konnte das größte PDF-exprimierende Neuron identifiziert werden. Dieses generierte Oszillationen, reagierte tagsüber aber nicht auf applizierte Lichtreize. Da es sich als sehr schwierig erwies PDF-Neurone mit der intrazellulären Methodik vollständig zu färben und physiologisch zu charakterisieren, wurden weitere Techniken praktiziert, um PDF-Neurone weiter analysieren zu können. Bei der Durchführung von Backfills in R. maderae aus dem kontralateralen optischen Stiel zusammen mit ultrananokristallinen Diamant- (UNCD) Filmen, Calcium-Imaging- Experimenten und immunchemischen Analysen konnte gezeigt werden, dass alle PDF-sensitiven kontralateral projizierten AME-Neurone durch PDF-Applikation inhibiert wurden, während alle ipsilateral verbleibenden Neurone durch PDF aktiviert wurden. Unter den PDF-sensitiven Neuronen waren auch kontralateral projizierende mittelgroße PDF-immunreaktive (ir) Neurone, die durch PDF inhibiert wurden und kleine lokale ipsilateral verbleibende PDF-ir-Neurone, die durch PDF aktiviert wurden. Diese PDF-exprimierenden Neurone hatten Autorezeptoren für ihr eigenes Peptid. Interessanterweise besaß das größte PDF-ir-Neuron, das in allen Zielbereichen der PDF-Zellen verzweigt, keine Autorezeptoren für sein eigenes Peptid.
