dcterms.abstract | Die Aminosäure-Sequenzierung an dem als "28 kDa-Thioredoxin f" beschriebenen Protein aus der Grünalge Scenedesmus obliquus hat gezeigt, dass dieses Protein mit dem als OEE bekannten Protein 1 aus dem Photosystem II identisch ist. Die früher postulierte Möglichkeit einer Fusion eines Thioredoxins mit einem Protein unbekannter Natur oder Insertion eines Thioredoxinfragments mit der typischen -Trp-Cys-Gly-Pro-Cys-Sequenz in ein solches Protein hat sich nicht bestätigt. Durch Anwendung einer auf das 33 kDa OEE-Protein ausgerichteten Präparationsmethode konnte gezeigt werden, dass das "28 kDa-Trx f" tatsächlich in den Thylakoidmembranen lokalisiert ist. Das Protein kann so innerhalb eines Tages in hoher Reinheit aus den Thylakoidmembranfragmenten eines Algenrohhomogenats isoliert werden; dabei bleibt die Fähigkeit des OEE-Proteins das chloroplastidäre Enzym Fructosebisphosphatase (FbPase) zu stimulieren erhalten. Mit gleichen Methoden wurden die Grünalgen Chlorella vulgaris und Chlamydomonas reinhardtii auf außergewöhnliche Proteine mit Trx-f Aktivität untersucht. Die hitze- und säurestabile Proteinfraktion aus Chlorella vulgaris enthält ein Protein mit vergleichbarer Molmasse von 26 kDa, das ähnlich wie in Scenedesmus eine Stimulation der chloroplastidären Fructosebisphosphatase zeigt. In dem hitze- und säurestabilen Proteinextrakt aus Chlamydomonas reinhardtii wird solche Aktivität nicht beobachtet. Eine Probe des rekombinanten, homogenen OEE-Proteins aus Spinat wurde auf Stimulation der chloroplastidären FbPase und NADPH-abhängigen Malatdehydrogenase (MDH) untersucht. Das Spinat OEE-Protein 1 zeigt mit diesen Enzymen keine Aktivität. Da das OEE-Protein 1 in Scenedesmus starke FbPase-Stimulation zeigt, die anderen Scenedesmus-Thioredoxine mit Molmassen von 12 kDa (Trx I und II) jedoch hohe Aktivität mit der zellulären Ribonucleotidreduktase zeigen, wird postuliert, dass das OEE-Protein die Funktion des Trx-f in vivo ersetzt. | eng |
dcterms.abstract | Amino acid sequencing of the 28 kDa-thioredoxin f (Trx f) protein from the green algae Scenedesmus obliquus exhibits that this protein is identical to the well-known oxygen-evolving enhancer (OEE) protein 1 of the photosystem II. The possibility of a fusion of a thioredoxin with a protein of unknown nature or insertion of a thioredoxin fragment with the typical -trp-cys-gly-pro-cys- sequence into such a protein, postulated previosly, was not confirmed. By application of a preparation method optimized for this 33 kDa OEE protein it could shown that the 28 kDa Trx f is located in the thylakoid membranes. In such a way the protein can be isolated from the thylakoid membranes of a crude extract of this algae in high purity within one day. The OEE protein 1 retains the ability to stimulate the chloroplast enzyme fructose bis-phosphatase (FbPase). Using the same methods the green algae Chlorella vulgaris and Chlamydomonas reinhardtii were examined for unusual proteins with Trx f activity. The heat and acid-stable protein fraction from Chlorella vulgaris contains a protein with comparable molecular mass of 26 kDa. This protein exhibits stimulation of the chloroplastic fructose bis-phosphatase similar to Scenedesmus protein. In heat and acid-stable protein fraction from Chlamydomonas reinhardtii no such activity is observed. A sample of recombinant, homogeneous OEE protein from spinach was examined for stimulation of the chloroplastic FbPase and NADPH dependent malate dehydrogenase (MDH). However the spinach OEE protein 1 does not show activity towards these enzymes. Since the OEE protein 1 shows strong FbPase stimulation in Scenedesmus, bat other Scenedesmus thioredoxins with molecular masses of 12 kDa (Trx I and II) show high activity with ribonucleotide reductase, it is postulated that the OEE protein replaces the function of Trx f in vivo. | eng |
