Microbial carbon use efficiency of litter with distinct C/N ratios in soil at different temperatures, including microbial necromass as growth component
(2022-08-10)
An incubation study was carried out to investigate the effects of litter quality, i.e. ¹⁵ N-labelled maize (C/N of 25.5) and Rhodes grass (C/N of 57.8) leaf litter on microbial carbon use efficiency (CUE) and priming effects in a moderate alkaline soil at two different temperatures (15 and 25 °C). CUE values were calculated from the isotopic composition of the particulate organic matter (POM) recovered as an index for the amount of non-decomposed litter. This approach allows the inclusion of microbial necromass growth ...

Development and Testing of the A1 Volumetric Air Sampler, an Automatic Pollen Trap Suitable for Long-Term Monitoring of eDNA Pollen Diversity
(2022-08-29)
Airborne pollen surveys provide information on various aspects of biodiversity and human health monitoring. Such surveys are typically conducted using the Burkard Multi-Vial Cyclone Sampler, but have to be technically optimized for eDNA barcoding. We here developed and tested a new airborne pollen trap, especially suitable for autonomous eDNA-metabarcoding analyses, called the A1 volumetric air sampler. The trap can sample pollen in 24 different tubes with flexible intervals, allowing it to operate independently in ...

The Thermodynamic Stability of Membrane Proteins in Micelles and Lipid Bilayers Investigated with the Ferrichrom Receptor FhuA
(2022-05-13)
Extraction of integral membrane proteins into detergents for structural and functional studies often leads to a strong loss in protein stability. The impact of the lipid bilayer on the thermodynamic stability of an integral membrane protein in comparison to its solubilized form in detergent was examined and compared for FhuA from Escherichia coli and for a mutant, FhuAΔ5-160, lacking the N-terminal cork domain. Urea-induced unfolding was monitored by fluorescence spectroscopy to determine the effective free energies ...

A comparison of online and live training of livestock farmers for an on-farm self-assessment of animal welfare
(2022-11-14)
One approach to strengthening the involvement of farmers or stockpersons in the evaluation and improvement of animal welfare is the implementation of an on-farm self-assessment. A valid comparison of the results with reference values, between or within farms, requires that training of the farmers and reliability testing have taken place. We investigated two different training methods (online vs. live) with a total of 146 livestock farmers from farms with dairy cows and calves, beef cattle, sows and suckling piglets, ...

Spatial and Temporal Variations of Stable Isotopes in Precipitation in the Mountainous Region, North Hesse
(2022-12-01)
Patterns of stable isotopes of water (¹⁸O and ²H) in precipitation have been used as tracers for analyzing environmental processes which can be changed by factors such as the topography or meteorological variables. In this study, we investigated the isotopic data in precipitation for one year in the low mountain range of North Hesse, Germany, and analyzed mainly for altitude, rainfall amount, and air temperature effects on a regional scale. The results indicate that the isotopic composition expressed an altitude ...

The potential of metabarcoding plant components of Malaise trap samples to enhance knowledge of plant-insect interactions
(2022-07-22)
The worldwide rapid declines in insect and plant abundance and diversity that have occurred in the past decades have gained public attention and demand for political actions to counteract these declines are growing. Rapid large-scale biomonitoring can aid in observing these changes and provide information for decisions for land management and species protection. Malaise traps have long been used for insect sampling and when insects are captured in these traps, they carry traces of plants they have visited on the body ...

Phospholipid fatty acids in soil—drawbacks and future prospects
(2021-12-21)
The current opinion and position paper highlights (1) correct assignation of indicator phospholipid fatty acids (PLFA), (2) specificity and recycling of PLFA in microorganisms, and (3) complete extraction and detection of PLFA. The straight-chain PLFA 14:0, 15:0, 16:0, and 17:0 occur in all microorganisms, i.e., also in fungi and not only in bacteria. If the phylum Actinobacteria is excluded from the group of Gram-positive bacteria, all remaining bacteria belong to the bacterial phylum Firmicutes, which should be ...

Mineralisation of distinct biogas digestate qualities directly after application to soil
(2020-11-02)
Biogas is an important energy source produced by the anaerobic fermentation of raw faecal slurries and plant residues. Separation of the total digestate increases the fertilizer quality of the liquid fraction and the carbon sequestration potential of the solid fraction. A 12-day incubation study was carried out to investigate the relationships between the chemical composition of different digestate qualities and the immediate response of soil microbial activity and biomass indices. The highest cumulative (Σ) CO2-C ...

Combined leaching and plant uptake simulations of PFOA and PFOS under field conditions
(2020-08-31)
Per- and polyfluoroalkyl substances (PFASs) are used in industrial production and manufacturing but were repeatedly detected in agricultural soils and therefore in cash crops in recent years. Dissipation of perfluoroalkyl acids (PFAAs), a sub-group of PFASs, in the environment was rather attributed to the formation of non-extractable residues (NER) than to degradation or transformation. Currently, there are no models describing the fate of PFAAs in the soil-plant continuum under field conditions, which hampers an ...
