Phylogenetische und morphologische Untersuchungen der Gattung Palifer (Hymenochaetales, Basidiomycota)

The Hyphodontia s.l. genus Palifer has only been phylogenetically examined by one available ribosomal ITS and LSU sequence from GenBank. This Palifer verecundus sequence indicates a position of Palifer among Xylodon. During the studies for the presented master thesis 15 new ITS and 11 LSU sequences have been generated including Palifer gamundiae, P. hjortstamii, P. wrightii, and P. verecundus. The most successful combination of primer pairs was ITS5/ITS2 and ITS3/ALR0 for ITS amplification. The LSU region was amplified by using the primer pairs LR0R/LR22 and LR22F/TW13. The forward primer LR22F was specifically designed to bind between D1 and D2 of the LSU region and proposed within this master thesis. The following phylogenetic analyses confirm Palifer as a synonym of Xylodon. Since the investigated species do not cluster within one clade it is inferred that not all Palifer species are closely related. Palifer gamundiae and P. hjortstamii could be nearly considered conspecific but have distinctive varying spore sizes. The genetic distances between Palifer verecundus-samples of different origins were found to be greater than between Palifer gamundiae and P. hjortstamii. The only Palifer wrightii sequence clusters near P. verecundus implying no relation to Hyphodontia s.s. species. In addition, the identity of two new species from Chile is discussed within this master thesis.
