Main soil microbial groups assessed by phospholipid fatty acid analysis of temperate alley agroforestry systems on crop- and grassland
(2024-01-12)
The phospholipid fatty acid (PLFA) composition of soils was analysed at three poplar-based silvo-arable systems and at one willow-based silvo-grassland alley agroforestry system in Central Germany. The objective was to analyse tree row effects on the PLFA composition of main fungal and main bacterial groups. The fungal groups were BAM (Basidiomycota + Ascomycota + Mucoromycota) and AMF (Arbuscular Mycorrhizal Fungi). The bacterial groups were Gram-negative, Firmicutes, and Actinobacteria. The total PLFA content varied ...

Lipidome remodeling in response to nutrient replenishment requires the tRNA modifier Deg1/Pus3 in yeast
(2023-10-21)
In the yeast Saccharomyces cerevisiae, the absence of the pseudouridine synthase Pus3/Deg1, which modifies tRNA positions 38 and 39, results in increased lipid droplet (LD) content and translational defects. In addition, starvation-like transcriptome alterations and induced protein aggregation were observed. In this study, we show that the deg1 mutant increases specific misreading errors. This could lead to altered expression of the main regulators of neutral lipid synthesis which are the acetyl-CoA carboxylase (Acc1), ...

Anthropogenic and natural disturbances increase local genetic diversity in an early spring geophyte (Ficaria verna Huds)
(2023-12-26)
The tetraploid Ficaria verna is a common spring geophyte in central Europe and is considered invasive in the USA and Canada. It is considered an almost seed-sterile taxon, relying on vegetative reproduction by underground tubers and aerial bulbils. Recent studies have revealed high levels of population genetic diversity in F. verna, raising the question of how genetic diversity is maintained and which factors may be responsible for the observed patterns. Polymorphic nuclear microsatellite markers were established to ...

Galactosamine and mannosamine are integral parts of bacterial and fungal extracellular polymeric substances
(2024-03-22)
Extracellular polymeric substances (EPS) are produced by microorganisms and interact to form a complex matrix called biofilm. In soils, EPS are important contributors to the microbial necromass and, thus, to soil organic carbon (SOC). Amino sugars (AS) are used as indicators for microbial necromass in soil, although the origin of galactosamine and mannosamine is largely unknown. However, indications exist that they are part of EPS. In this study, two bacteria and two fungi were grown in starch medium either with or ...

Optimization of the bicinchoninic acid assay for quantifying carbohydrates of soil extracellular polymeric substances
(2023-12-23)
Background and aims
The bicinchoninic acid (BCA) method was not yet applied on soil extracts of extracellular polymeric substances (EPS) to quantify polysaccharides, although this might be possible by introducing a cleavage step to produce monosaccharides. A pre-extraction with CaCl₂ to remove interfering substances is usually performed before extracting EPS. The main objective of this study was to optimize the BCA assay for total carbohydrates quantification by applying a hydrolysis step to the EPS extracts, while ...
