Loss of Anticodon Wobble Uridine Modifications Affects tRNALys Function and Protein Levels in Saccharomyces cerevisiae
(2015)
In eukaryotes, wobble uridines in the anticodons of tRNALysUUU, tRNAGluUUC and tRNAGlnUUG are modified to 5-methoxy-carbonyl-methyl-2-thio-uridine (mcm5s2U). While mutations in subunits of the Elongator complex (Elp1-Elp6), which disable mcm5 side chain formation, or removal of components of the thiolation pathway (Ncs2/Ncs6, Urm1, Uba4) are individually tolerated, the combination of both modification defects has been reported to have lethal effects on Saccharomyces cerevisiae. Contrary to such absolute requirement ...

Loss of wobble uridine modification in tRNA anticodons interferes with TOR pathway signaling
(2014)
Previous work in yeast has suggested that modification of tRNAs, in particular uridine bases in the anticodon wobble position (U34), is linked to TOR (target of rapamycin) signaling. Hence, U34 modification mutants were found to be hypersensitive to TOR inhibition by rapamycin. To study whether this involves inappropriate TOR signaling, we examined interaction between mutations in TOR pathway genes (tip41Δ, sap190Δ, ppm1Δ, rrd1Δ) and U34 modification defects (elp3Δ, kti12Δ, urm1Δ, ncs2Δ) and found the rapamycin ...

Microsatellites from Fosterella christophii (Bromeliaceae) by de novo transcriptome sequencing on the Pacific Biosciences RS platform
(2016)
• Premise of the study: Microsatellite markers were developed in Fosterella christophii (Bromeliaceae) to investigate the genetic diversity and population structure within the F. micrantha group, comprising F. christophii, F. micrantha, and F. villosula. • Methods and Results: Full-length cDNAs were isolated from F. christophii and sequenced on a Pacific Biosciences RS platform. A total of 1590 high-quality consensus isoforms were assembled into 971 unigenes containing 421 perfect microsatellites. Thirty primer sets ...

Signaling of Pigment-Dispersing Factor (PDF) in the Madeira Cockroach Rhyparobia maderae
(2014)
The insect neuropeptide pigment-dispersing factor (PDF) is a functional ortholog of vasoactive intestinal polypeptide, the coupling factor of the mammalian circadian pacemaker. Despite of PDF's importance for synchronized circadian locomotor activity rhythms its signaling is not well understood. We studied PDF signaling in primary cell cultures of the accessory medulla, the circadian pacemaker of the Madeira cockroach. In Ca2+ imaging studies four types of PDF-responses were distinguished. In regularly bursting type ...

Development of 18 Polymorphic Microsatellite Markers for Vinca minor (Apocynaceae) via 454 Pyrosequencing
(2015)
• Premise of the study: Polymorphic microsatellite markers were developed in Vinca minor (Apocynaceae) to evaluate the level of clonality, population structure, and genetic diversity of the species within its native and introduced range.
• Methods and Results: A total of 1371 microsatellites were found in 43,565 reads from 454 pyrosequencing of genomic V. minor DNA. Additional microsatellite loci were mined from publicly available cDNA sequences. After several rounds of screening, 18 primer pairs flanking di-, tri-, ...

How do consumers search for and process corporate social responsibility information on food companies’ websites?
(2019-01-03)
The importance of corporate social responsibility (CSR) communication for food companies has grown substantially ever since global action plans such as the UN Sustainable Development Goals have begun targeting topics which are highly relevant to food companies. Furthermore, various food scandals have caused consumers to question current food production methods, leading to a search for more information on ethical production methods. However, it is not known how consumers search for and process information on websites ...

Human and farm influences on dairy cows' responsiveness towards humans - a cross-sectional study
(2018-12-31)
The human-animal relationship can have a major impact on cow welfare and stockpersons´ work safety and quality. This cross-sectional study investigated possible effects of stockperson and farm related factors on cows´ behaviour towards humans in different test situations on a range of farm types including farms with automatic milking system. On 32 dairy farms, cows´ avoidance distances (AD), tolerance to tactile interactions (TTI), behavioural reactions during and after release from restraint (RB) and expressive ...

Amplified Spontaneous Emission in Paper
(2019-02-12)
Industrially produced copy paper is presented as a novel optical material for narrow-band stimulated emission. Fluorescent brightening agents (FBA) such as Calcofluor White provide enough gain to exceed the threshold for amplified spontaneous emission. By applying an additional dye such as Coumarin 307, or simply a highlighter pen, the emission line can be shifted from blue (~440 nm), towards turquoise (>480 nm), up to orange (590 nm) which can be useful for spectroscopic applications. These properties are demonstrated ...

Participative Processes as a Chance for Developing Ideas to Bridge the Intention-Behavior Gap Concerning Sustainable Diets
(2018-11-27)
Sustainable diets are drivers and results of sustainable food systems. Therefore, they are crucial for improving our global diet-related problems. When trying to adopt sustainable diets, people often struggle with the gap between their good intentions and their actual behavior. Here we see a need for support. To understand people’s needs and what could help them, it stands to reason that they can be directly involved in the development processes for appropriate ideas. On that account, we conducted six workshops in ...

Measuring Product Material Footprint as New Life Cycle Impact Assessment Method: Indicators and Abiotic Characterization Factors
(2019-04-02)
The global economy is using growing amounts of natural resources such as raw materials, water, and land by making and using goods, services, and infrastructure. Aspirations on international, regional, and national levels e.g., the Sustainable Development Goals, the EU flagship initiative Roadmap to a Resource Efficient Europe or the German Program for Resource Efficiency are showing an urgent need to bring the global raw material use down to sustainable levels. An essential prerequisite to identify resource efficient ...
